When working in SQL Server Reporting Services it is easy to create the Data Source connection using the wizards in Report Designer (Visual Studio) or Report Builder.

Connection Wizard

However, if you need to create a new data source within Report Manager, no wizard is provided.

Report Manager New Data Source

If you can, it is simple enough to open a different connection manager that uses the same a similar data source.  However, this may not be available if this is a new server, you lack permissions or the target is a new server type.  Here is a handy link you can bookmark that provides you with the connection string formats.
